This study aims to evaluate the safety, acceptability, and pharmacokinetics of an investigational tenofovir-based rectal douche for on-demand HIV prevention. Many individuals who engage in receptive anal intercourse use rectal douches as part of their preparation for sex. The primary objectives of this study are to assess the safety, acceptability, and evaluate the drug concentrations achieved with the rectal douche compared to daily oral PrEP tablets. This study will also determine participants' preferences between the two methods and help establish whether the tenofovir rectal douche can be a viable on-demand option for HIV prevention. 

Eligibility: 

  • ≥ 18 Years Old  
  • Male or Transgender Woman
  • HIV Negative
  • Had receptive anal sex (Bottom) at least once in the past 3 months.
  • No PrEP for past 8 weeks.
